COPII-coated vesicle formation reconstituted with purified coat proteins and chemically defined liposomes
Article Abstract:
The effects of binding between COPII coat proteins and liposomes were investigated using ultrastructural analysis. Different combinations of COPII proteins were incubated with microsomal membranes enriched with endoplasmic reticulum to examine the mechanism of COPII vesicle formation. Results show that the binding of COPII coat proteins to liposomes induces coated patches, coated vesicles and coated buds. It was found that the assembly of the COPII coat on the endoplasmic reticulum promotes the budding of COPII-coated vesicles.

The small GTPase Sar1p directly initiates membrane curvature during vesicle biogenesis. The initiation of vesicle budding by Sarl1p, which is a COPII protein, couples the generation of membrane curvature with coat-protein assembly and cargo capture.
