Cloning and expression of a phloretin hydrolase gene from Eubacterium ramulus and characterization of the recombinant enzyme
Article Abstract:
The cloning and sequencing of the encoding gene and the subsequent purification and characterization of the recombinant phloretin hydrolase of Eubacterium ramulus expressed in Escherichia coli is described. The strong inhibition of phloretin hydrolase by the metal-ion-chelating agent o-phenanthroline reveals that metal ions, such as iron may be involved in catalysis and the products of phloretin hydrolysis reduce the activity of the E. ramulus phloretin hydrolase, while the enzyme from E. herbicola was not inhibited by these compounds even at higher concentrations. It is concluded that further research will be carried out to unravel the mechanism underlying the hydrolysis of phloretin.

Degradation of quercetin and luteolin by Eubacterium ramulus

Eubacterium ramulus may contribute to the degradation of flavonoids in the human diet. This bacterium resides in the human gastrointestinal tract. The metabolism of flavonoids proceeds via different reactions depending on the presence of a 3-hydroxyl group on the flavonoid.

Anaerobic degradation of flavonoids by Clostridium orbiscindens

Research demonstrates that the quercetin-degrading isolates from human fecal matter are Clostridium orbiscindens, which anaerobically degrade flavonoids such as quercetin, taxifolin, luteolin, eriodictyol, apigenin and others.
