Cloning of the spoT gene of "Candidatus phlomobacter fragariae" and development of a PCR-restriction fragment length polymorphism assay for detection of the bacterium in insects
Article Abstract:
Researchers describe a PCR test for the spoT gene from proteobacteria that can be used to identify which insects are vectors for the proteobacterium "Candidatus phlomobacter fragariae". Using this test, they found that whiteflies could carry the bacterium.

Stable transformation of the Xylella fastidiosa citrus variegated chlorosis strain with oriC plasmids
Article Abstract:
Researchers used a plasmid to introduced a gene for kanamycin resistance into Xylella fastidiosa, which causes citrus variegated chlorosis. This successful integration means that this technique can be used to inactivate specific bacterial genes.

Engineering of a single-chain variable-fragment (scFv) antibody specific for the stolbur phytoplasma (Mollicute) and its expression in Escherichia coli and tobacco plants
Article Abstract:
The scFv gene, which is specific for the stolbur phytoplasma mollicute, was cloned and expressed in Escherichia coli cells and tobacco plants. The mollicutes were found to be confined to the sieve tubes within the phloem tissues. However, scFv was not detected in tobacco protein extracts by Western blotting. One transgenic tobacco line, 1A6, was chosen for challenge inoculation with the stolbur phytoplasma. Transgenic tobacco shoots grafted on a stolbur phytoplasma-infected tobacco rootstock, grew without any symptoms while normal tobacco shoots had the symptoms and eventually died.
