Conditions for vigorous growth on sulfide and reactor-scale cultivation protocols for the thermophilic green sulfur bacterium Chlorobium tepidum
Article Abstract:
Protocols for the cultivation of Chlorobium tepidum in a 22-liter photobioreactor under optimal conditions have been developed. The protocols enumerate the conditions required for the green sulfur bacterium to thrive with sulfide as the sole electron source. The bacterium is notorious for its inability to grow on sulfide alone. C. tepidum could be grown at rates and final cell yields comparable to that with thiosulfate if the sulfide concentration is maintained below 0.1 mM and the culture redox potential is at -300 at plus or minus 20 mV.

Chromosomal gene inactivation in the green sulfur bacterium Chlorobium tepidum by natural transformation
Article Abstract:
Three antibiotic resistance markers have been found for Chlorobium tepidum that would be useful for gene inactivation: a spectinomycin-streptomycin marker, a gentamicin marker, and an erythromycin marker. The 2.15-Mb genome of the green sulfur bacterium has been sequenced and contains about 2,284 open reading frames.

Reactor-scale cultivation of the hyperthermophilic methanarchaeon Methanococcus jannaschii to high cell densities
Article Abstract:
Research conducted on the cultivation of Methanococcus jannaschii in a reactor to demonstrate media recipes and protocols for its mass culture is analyzed. Results indicate the possibility of scaling the cultures up to higher volumes.
