Evaluation of the DNA fingerprinting method AFLP as a new tool in bacterial taxonomy
Article Abstract:
AFLP fingerprinting method can be used on any bacterial genomic DNA, irrespective of its complexity or origin. The DNA fragments formed by digestion with restriction enzymes are ligated to restriction halfsite-specific adaptors. These adaptors form primer-binding sites that allow amplification of the fragments. Radioactively labeled selective primers produce banding patterns that are processed to identify band-based similarity coefficients. AFLP combines the features of PCR-based technology and RFLP, and has high reproducibility and resolution.

Partial small subunit rRNA gene sequence analysis and limited phenotypic analyses are used for exploring diversity of a collection of lactococcus isolates including Lactococcus lactis isolates of dairy and nondairy origin. The pheS DNA sequence analysis has yielded two genetic groups that are dissimilar to the other genotyping analysis-based lineages, which has shown a disparate acquisition route for this gene.
