FAN, a novel WD-repeat protein, couples the p55 TNF-receptor to neutral sphingomyelinase
Article Abstract:
A new protein, which attaches with the neutral sphingomyelinase (N-SMase) of the human tumor necrosis factor-R55, is identified by yeast interaction trap system and a cellulose-bound peptide scanning library. The protein FAN or factor associated with N-SMase activation is identified by sequence analysis as a WD-repeat protein, which are active in signal transduction. Overexpression further reveals that FAN helps in the activation of N-SMase by tumor necrosis factor (TNF). This suggests its possible relevance in controlling key inflammatory cellular reactions to TNF.

Mice deficient for the 55kd tumor necrosis factor receptor are resistant to endotoxic shock, yet succumb to L. monocytogenes infection
Article Abstract:
A mutant mouse strain that is deficient in the 55 kd receptor of the tumor necrosis factor (TNFRp55) was established. The splenocytes of TNFRp55 -/- mice express normal numbers of high affinity TNFRp75, indicating that the expression of TNFp75 is independent of TNFp55. Moreover, major alterations with respect to TNF signaling and host responses to specific stimuli were demonstrated. The findings suggest that TNFRp55 mediated signals play a role in both toxic and protective effects of TNF in vivo.

Impaired negative selection of T cells in Hodgkin's disease antigen CD30-deficient mice
Article Abstract:
The thymus of mice lacking antigen CD30 receptors have increased amounts of thymocytes. Killing of thymocytes by CD3 cross-linking was ineffective both in vivo and in vitro. Breeding the CD30 mutation separately into alpha-beta-TCR- or alpha-delta-TCR-transgenic mice revealed a substantial impairment of negative but not in positive selection of T cells. Findings indicated that the CD30 receptor is implicated in cell death signaling.
