Group-specific 16S rRNA-targeted oligonucleotide probes to identify thermophilic bacteria in marine hydrothermal vents
Article Abstract:
Group-specific 16S rRNA-targeted oligonucleotide probes were developed and characterized for their ability to detect four types of thermophilic bacteria in deep-sea hydrothermal/marine coastal environments. The probes include species of the genus Thermus, the genera Thermatoga and Thermosipho, the thermophilic members of the Bacillus genus and Aquificales order. Results reveal that probes for various phylogenetic levels may be employed in hydrothermal vent microbial ecology research.

Distribution of microorganisms in deep-sea hydrothermal vent chimneys investigated by whole cell hybridization and enrichment culture of thermophilic subpopulations
Article Abstract:
The distribution of microorganisms in deep-sea hydrothermal vent chimneys was investigated using a whole-cell hybridization and enrichment culture of thermophilic subpopulations. The diversity of the thermophilic microbial community was assessed by using a combination of traditional culture-based enrichments. Results show that whole-cell hybridization is based on phylogenetic staining. Furthermore, culture-based enrichment is dependent on the culturability of cells.

Thermophilic lifestyle for an uncultured archaeon from hydrothermal vents: Evidence from environmental genomics
Article Abstract:
A comparative analysis of two genome fragments isolated from a diverse and widely distributed group of uncultured euryarchaea from deep-sea hydrothermal vents is presented. It shows that optimal activity and thermostability of a DNA polymerase predicted in one fragment are close to that of the thermophilic acrhaeon Thermoplasma acidophilum, providing evidence for a thermophilic way of life of this group of uncultured archaea.
