Hotspots for unselected Ty1 transposition events on yeast chromosome III are near tRNA genes and LTR sequences
Article Abstract:
Yeast strains with single marked Ty1 insertions on chromosome III were constructed and the insertions were monitored by pulsed field gel electrophoresis. The insertions were done randomly, 32 of them being cloned by the PCR technique by which the DNA sequences were derived. The experiment reveals that the genome regions, mapped by the Ty1 integration apparatus, do not contain any special DNA sequences. Since the DNA sequences also do not cooperate for the Ty1 integration in vitro, the targeted regions possess special characteristics facilitating specific recognition in vivo.

Visualizing the spatial relationships between defined DNA sequences and the axial region of extracted metaphase chromosomes
Article Abstract:
In mammalian chromosomes, the DNA strand forms looped structures around a central scaffold during metaphase. The scaffold shows up as a brightly colored region on stained photographs of hybridization. This scaffold has been identified as the centromere, and specific DNA regions are always associated with this axis. These regions have been identified as those involved in replication. Meanwhile, regions coding for ribosomal DNA are also found to be closely associated with the chromosome axis.

Cohesins bind to preferential sites along yeast chromosome III, with differential regulation along arms versus the centric region
Article Abstract:
A methodology was created to analyze the distribution of cohesins Mcd1/Scc1 and Smc1 along yeast chromosome III. The methodology, which involves the use the use of an extended chromatin immunoprecipitation, can also be utilized in analyzing the distribution of any DNA-binding protein. It was revealed that cohesins Mcd1/Scc1 and Smc1 occur preferentially at the same approximately 23 positions. Intense signals were obtained from sites in an approximately 50 kb region around the centromere.
