Identity elements in tRNA-mediated transcription antitermination: implication of tRNA D- and T-arms in mRNA recognition
Article Abstract:
Identity elements in tRNA-mediated transcription antitermination have been studied with attention to implications of tRNA D- and T-arms in mRNA recognition. The mRNA-tRNA interaction involves parts of the tRNA in addition to the anticodon and the acceptor in an earlier model. The additional interactions may contribute to the recognition of a specific tRNA, and thus to the specificity and efficacy of the regulatory response.

Research presented concerns gene regulation in Gram-positive bacteria, focusing on how tRNA-mediated genetic transcription antitermination regulates the expression of several operons involved in amino acid biosynthesis and aminoacyl-tRNA-synthetase-encoding genes. A model of these operons and genes is presented.

Transcription of the trp operon in Lactococcus lactis is controlled by antitermination in the leader region
Article Abstract:
A mutagenesis analysis of the Lactococcus lactis was performed to characterize the regions of trp leader needed to repress the transcription of the trp operon in the presence of tryptophan. A screen was created to allow the isolation of deregulated trp mutants. Mutations affecting the function of the transcription terminator T1 exhibiting the functional features of prokaryotic intrinsic transcription terminators were exhibited. A first class of mutants containing short deletions or point mutations that affect the T1 stem was also observed.
