Induction of apoptotic program in cell-free extracts: requirement for dATP and Cytochrome c
Article Abstract:
The established cell-free system based on cytosols of normally growing cells reproduces aspects of the apoptotic program in vitro. The addition of dATP initiates the apoptotic program. A 15 kDa protein necessary for in vitro apoptosis was yielded via cytosol fractionation. An analysis of the absorption spectrum and protein sequence showed that the protein is cytochrome c. Apoptotic activity was reduced when cytochrome c was removed from cytosol via immunodepletion or when sucrose was added to stabilize mitochondria during cytosol preparation.

Bid, a Bcl2 interacting protein, mediates cytochrome c release from mitochondria in response to activation of cell surface death receptors
Article Abstract:
An experiment was conducted to purify a cytosolic protein that induces cytochrome c release from mitochondria in response to an apical caspase activated by cell surface death receptors. Peptide mass fingerprinting showed that the protein is Bid, a BH3 domain-containing protein that interacts with either BcL2 and Bax. Further analysis involving mutation at the BH3 domain showed that Bid relays an apoptotic signal from the cell surface to the mitchondria.

DFF, a heterodimeric protein that functions downstream of caspase-3 to trigger DNA fragmentation during apoptosis
Article Abstract:
Researchers have identified and purified from HeLa cytosol a protein that induces DNA fragmentation in coincubated nuclei after activation by caspase-3. The protein, known as DNA Fragmentation Factor, is a heterodimer of 40kDa and 45 kDa subunits. Caspase-3 divides the 45 kDa subunit at two sites to create an active factor that induces DNA fragmentation without further need for caspase-3 or other cytosolic proteins.
