Kinetics of cellulose digestion by Fibrobacter succinogens S85
Article Abstract:
Fibrobacter succinogens S85 is an active cellulolytic bacteria, but cell extracts and nongrowing cells are incapable of producing crystalline cellulase activity. Cells growing exponentially on cellobiose initiated cellulose digestion and succinate production immediately and can be used as an index of enzyme activity against crystalline cellulose. Cells incubated with cellulose fail to produce detectable cellobiose, while those that are preincubated for a short time with thiocellobiose lose their ability to digest cellulose.

Cloning, sequencing, and expression of a Thermomonospora fusca protease gene in Streptomyces lividans
Article Abstract:
The sequencing of the Thermomonospora fusca YX extracellular protease gene cloned into Escherichia coli and Streptomyces lividans shows that the gene is an open reading frame. The gene encodes a molecule containing a signal sequence, an N-terminal prosequence and a 194-mature protease belonging to the chymotrypsin family. S. lividans expressing the gene secretes the protease, which is bound to an extracellular protease inhibitor.

Article Abstract:
The processivity, substrate binding and mechanism of cellulose hydrolysis are discussed. Processivity was controlled by residues in the catalytic cleft.
