Phylogenetic probes for analyzing abundance and spatial organization of nitrifying bacteria
Article Abstract:
Phylogenetic hierarchical 16S rRNA oligonucleotide probes specific for ammonia- and nitrite-oxidizing bacteria are suitable for the identification of bacteria in environmental samples by slot blot, whole-cell or in situ hybridization. The probes are specific for Proteobacteria beta subclass, Nitrosomonas, Nitrosovibrio-Nitrosolobus-Nitrosospira and Nitrobacter species. In situ hybridization results show that biofilm and nitrifying activated sludge samples contain mainly Nitrosomonas bacteria. Nitrosomonas bacteria are found near and sometimes associated with Nitrobacter bacteria.

Oligonucleotide microchips as genosensors for determinative and environmental studies of microbiology
Article Abstract:
The applicability of parallel hybridization of environmental nucleic acids to many oligonucleotides immobilized in a matrix of polyacrylamide gel pads on a glass slide was investigated. Oligonucleotides complementary to small-subunit rRNA sequences of selected microbial groups, encompassing key genera of nitrifying bacteria, were found to selectively retain labeled target nucleic acid derived from either DNA or RNA forms of the target sequences.

Critical evaluation of membrane supports for use in quantitative hybridizations
Article Abstract:
The Magna Charge and Immobilon-N membranes used in the oligonucleotide probe hybridization of 16S rRNA give higher levels of detectability than the MagnaGraph, Magna, and Nytran membranes. The difference in the signal response of the membranes is between 10 to 50%. The Magna Charge and Immobilon-N membranes have the smallest signal response variability. The membranes provide supports for nucleic acid immobilization during hybridization.
