Plasposons: modular self-cloning minitransposon derivatives for rapid genetic analysis of gram-negative bacterial genomes
Article Abstract:
The TnMod is a basic plasposon developed for use in the rapid genetic analysis of gram-negative bacterial genomes. It combines features of mini-transposons and self-cloning transposons to allow for the cloning and mapping of DNA flanking its insertion site. The plasposon was constructed by combining Tn5 inverted repeats, a conditional origin of replication, exchangeable antibiotic resistance cassettes and rare restriction endonuclease multiple cloning sites in a modular arrangement. These features make the TnMod ideal for mapping insertional mutations from bacteria.

Cloning, characterization, and nucleotide sequence of a gene encoding Microbispora bispora BglB, a thermostable beta-glucosidase expressed in Escherichia coli
Article Abstract:
The Microbispora bispora bglB gene codes for a thermostable beta-glucosidase which is also resistant to end product inhibition, making it an attractive candidate for industrial use. The M. bispora bglB gene was cloned and expressed in Escherichia coli. The results showed that bglB codes for a 52-kDa enzyme which was actually activated by glucose concentrations from two to five percent. Sequence analysis and comparisons revealed conserved regions within the N- and C-terminal domains which may be involved in binding or catalysis.
