Role of phosphoinositide 3-OH kinase in cell transformation and control of the actin cytoskeleton by Ras
Article Abstract:
The role of phosphoinosidyl kinase (PI 3-kinase) on the Ras transformation of cells was investigated by analyzing the effect of inhibition of the dominant-negative mutant of the enzyme's regulatory subunit, p85, on V12 Ras transformation of NIH 3T3 cells. Inhibition of growth of Ras-transformed NIH 3T3 cells infected with retro viruses expressing dominant-negative p85 was noted. Furthermore, the effect of PI3-kinase on Ras-induced cytoskeletal reorganization was determined by using porcine aortic endothelial (PAE) cells. Activated mutant PI 3-kinase induced cortical actin rearrangement and membrane ruffling on Ras-activated PAE cells.

The actin cytoskeleton and lymphocyte activation
Article Abstract:
Actin cytoskeletons are emerging as an integral part of lymphocyte activation and the hypothesis that actin cytoskeleton could be involved in intra-cell signalling was first made in the 1970s. Most steps in intra-cell signaling can be regulated by induced proximity in specific architectures. Thus actin-dependent clustering of signaling molecules could well serve the purpose of controling effective molarity. Future understanding of the actin cytoskeleton involvement in lymphocyte activation will likely rely on the identification of certain, defective biochemical pathways.

Identification of a nef allele that causes lymphocyte activation and acute disease in macaque monkey
Article Abstract:
The YE variant of nef allele replicates well in resting peripheral blood mononuclear cell cultures and causes extensive T lymphocyte activation which results in an acute disease in rhesus and pig tailed monkeys. The disease is characterized by severe diarrhea, rash and extensive lymphoid proliferation in the gastrointestinal tract. Both 239nef and YEnef are found to combine with src in cotransfected COS cells, and both 60 kDa src and 34 kDa are phosphorylated at tyrosine in these cells. Furthermore, the YEnef gene transformed NIH 3T3 cells in culture.
