Article Abstract:
DNA target-site recognition rules for a lactococcal group II intron make possible retargeting of the intron to certain DNA sequences. DNA target-site requirements for the Lactococcus lactis L1.LtrB group II intron in vivo and in vitro. Results indicate a model like yeast mtDNA introns. The L1LtrB intron can be changed to insert at new sites in a plasmid-borne thyA gene in E. coli.

The potential involvement of cellular factors of Escherichia coli mutants in retrohoming of the Lactococcus lactis intron is examined. The completion of retrohoming is viewed as a DNA repair process, with features that may be shared by other non-LTR retroelements.

Retrohoming of a bacterial group II intron: mobility via complete reverse splicing, independent of homologous DNA recombination
Article Abstract:
Ll.LtrB, a mobile group II intron of Lactococcus lactis, was utilized to examine the homing pathway in genetically tractable bacterial systems. It was proven that the mobility of Ll.LtrB can be attributed to an RNA-based retrohoming mechanism in both L. lactis and Escherichia coli. The results can be used to develop a model wherein intron integrates into a recipient DNA and functions as a template for cDNA synthesis.
