Site-specific pseudouridine formation in preribosomal RNA is guided by small nucleolar RNAs
Article Abstract:
The mechanisms that mediate the recognition of appropriate uridines for pseudouridine formation were analyzed during the nucleolar maturation of eukaryotic ribosomal RNAs. Analysis of ribosomal signal sequences that direct the site-specific pseudouryidylation of mammalian ribosomal RNA indicated the role of the yeast box H/ACA snoRNAs in guiding the RNA during site-specific pseudouridylation. Furthermore, site-specific pseudouridylation of yeast RNA was inhibited by the depletion of snR5 and snR36.

Site-specific ribose methylation of preribosomal RNA: a novel function for small nucleolar RNAs
Article Abstract:
Genetic depletion and reconstitution studies on yeast U24 antisense small nucleolar (sno)RNAs reveal that fibrillarin-associated snoRNAs behave as guide RNAs in the site-specific 2'-O-methylation of preribosomal RNA. The antisense elements along with the D and D' box of the snoRNA supply the necessary information for selecting the target nucleotides for the methylation reaction. Eukaryotic RNAs thus have a more diverse function than believed.

Small nucleolar RNAs direct site-specific synthesis of pseudouridine in ribosomal RNA
Article Abstract:
The synthesis of Psi in small nucleolar RNA (snoRNA)-depleted yeast cells and snoRNA sequences was analyzed to characterize the role of snoRNAs in cellular Psi formation. Analysis of Psi formation in yeast cells that were depleted with snoRNAs indicated the role of ribosomal RNA-snoRNA base pairing in the formation of Psi. Furthermore, snoRNAs that belong to the ACA family mediated site selection for the synthesis of Psi in yeast cells.
