Targeted mutagenesis of Tsix (ital) leads to nonrandom X inactivation
Article Abstract:
Targeted mutagenesis of Tsix(ital) is discussed. This phenomenon has been shown to lead to nonrandom X inactivation. A targeted deletion of Tsix(ital) in female and male mouse cells was created. With deficiency of Tsix(ital) RNA, X chromosome counting stays intact with female cells inactivating one X and male cells blocking X inactivation. But heterozygous female cells had skewed Xist(ital) expression and primary nonrandom inactivation of the mutant X. Ability of mutant X to block Xist(ital) accumulation is compromised.

Genomic imprinting disrupted by a maternal effect mutation in the Dnmt1 gene
Article Abstract:
Genomic imprinting has been found to be disrupted by a maternal effect mutation in the Dnmt1 gene. Dnmt1o was eliminated by deletion of the oocyte-specific promoter and first axon from the Dnmt1 locus. Homozygous offspring were normal, but most heterozygous fetuses of homozygous females died in later gestation. It appears that the Dnmt1o variant of Dnmt1 gives maintenance methyltransferase activity of imprinted loci in the fourth embryonic S phase specifically. .

Disruption of imprinted X inactivation by parent-of-origin effects at Tsix
Article Abstract:
The antisense Xist gene, Tsix, has been found to control imprinting. Tsix is expressed maternally, and mice with a Tsix deletion have normal paternal transmission, but maternal transmission is impaired. An imprinting center at Tsix is proposed. Disruption of imprinted X inactivation by parent-of-origin effects at Tsix is discussed.
