Adaptation to culture of human embryonic stem cells and oncogenesis in vivo
Article Abstract:
The karyotypic changes observed in culture adaptation, the parallels between adaptation and germ cell tumorigenesis and the implications of this process in the maintenance and therapeutic use of human embryonic stem cells (HESCs) are discussed. Alteration were found in mitochondrial DNA in late-passage HESC lines cultured under high oxygen tension, implying that the high oxygen tension could be responsible for promoting chromosome abnormalities.

Derivation of human embryonic stem cells in defined conditions
Article Abstract:
The cryptic art of cell culture has proved a major stumbling block on human embryonic stem (hES) cells. The feeder-independent hES cell culture that includes protein components solely derived from recombination sources or purified from human material was reported describing the derivation of two new hES cell lines in these culture conditions.

Recurrent gain of chromosomes 17q and 12 in cultured human embryonic stem cells
Article Abstract:
A selective advantage for the propagation of undifferentiated human embryonic stem (hES) cells is provided by the increased dosage of chromosome 17q and 12 gene(s). These findings in the hES cells are informative for transplantation therapies in which the use of aneuploid cells could be determined.
