Human C5aR knock-in mice facilitate the production and assessment of anti-inflammatory monoclonal antibodies
Article Abstract:
Neutrophils from human C5aR knock-in mice, in which the mouse C5aR coding region is replaced with that of human C5aR is used to immunize wild-type mice and to generate high-affinity antagonist monoclonal antibodies (mAbs) to human C5aR. Results reveal that of ~40 mAbs generated to C5aR, all potent inhibitors recognized a small region of the second extracellular loop that seems to be critical for regulation of receptor activity.

Rapid generation of specific antibodies by enhanced homologous recombination
Article Abstract:
The rapid generation of specific monoclonal antibodies using the chicken DT40 B-cell line undergoing gene conversion is reported. It is discovered that the gene conversion frequency at the immunoglobulin locus is increased by treating DT40 cells with a histone deacetylase inhibitor, trichostatin A, thereby generating diversity at the immunoglobulin locus in the majority of treated cells.

Simultaneous targeting of multiple disease mediators by a dual-variable-domain immunoglobulin
Article Abstract:
A generally applicable model of a dual-specific, tetravalent immunoglobulin G (IgG)-like molecule (dual-variable-domain Ig (DVD-Ig)) is developed, which is engineered from two monoclonal antibodies while preserving activities of parental antibodies. The studies of DVD-Ig protein in an animal disease model has shown that it can be used as therapeutics in human diseases.
