Article Abstract:
A demonstration that transgene sensitization of immune cells could be prevented using a component of the RNA interference (RNAi) machinery is presented. By tagging the trasngene with a sequence complementary to a microRNA (miRNA) that is specific to cells of the blood lineage, it is shown that immune system priming could be circumvented.

Isotype selection in antibody engineering
Article Abstract:
Study shows that antibodies of the IgG4 subclass exchange light chains in vivo, yielding bispecific products are likely to have unexpected effects in a therapeutic use. The most suitable isotype must be selected for designing antibodies for therapeutic use after considering the distinct properties of the specific isotypes.

An immunoglobulin G (IgG) variant that blocks FcRn function is described by binding tightly to the receptor and competing with endogenous IgGs, thereby hastening their degradation. IgG is the longest-lived and most highly expressed of the serum proteins, as befits its major functions in immunity and immunoregulation.
