Simultaneous binding of minor groove binder and intercalator to dodecamer DNA: Importance of relative orientation of donor and acceptor in FRET
Article Abstract:
The steady-state picosecond time-resolved fluorescence and polarization gated anisotropy are used to establish simultaneous binding of an intercalator (ethidium bromide) and a minor groove binder (Hoeschst33258, H258) to a dodecamer DNA of specific sequence. The Forster resonance energy transfer (FRET) studies have established that the value of the orientation factor should be reconsidered for the correct estimation of the donor-acceptor distance when the ligands are simultaneously bound to a specific region of biological macromolecules.

Spectroscopic studies on ligand-enzyme interactions: complexation of [alpha]-chymotrypsin with 4',6-diamidino-2-phenylindole (DAPI)
Article Abstract:
The interaction of two structurally related proteolytic enzymes trypsin and [alpha]-chymotrypsin (CHT) with 4',6-diamidino-2-phenylindole (DAPI) is studied by using steady-state and picosecond time-resolved spectroscopic methods. The differences found in the interactions of two structurally similar enzymes with the same molecule can be used in the design of specific mimics or inhibitor of the enzymes.

Dynamics in the DNA recognition by DAPI: exploration of the various binding modes

The dynamics of solvation is used for exploring the binding interaction of 4',6-diamidino-2-phenylindole (DAPI) to DNA oligomers of different sequences. The analysis has explored the structure-dynamics correlation of the DAPI-DNA complex in the two distinct modes of interaction of DAPI with DNA.
