Old approach yields new antibiotic
Article Abstract:
A new antibacterial compound with an astoundingly nefarious mechanism of action is introduced revealing that compound acts by removing the stringent controls on a bacterial protease, ClpXP, unleashing the enzyme to degrade cellular proteins in an unregulated manner. The parent compound was originally isolated from the culture broth of Streptococcus hawaiiensis, highlighting the value of natural product compounds and of mining the patent literature.

Multi-targeting by monotherapeutic antibacterials
Article Abstract:
A study discusses the distinguishing quality of the existing targets of monotherapeutic antibacterials, with their low potential for rapid endogenous resistance development that appears to be based on their capacity to act at multiple targets. It is suggested that ensuring multiple drug-target interactions, whether intra- or intermolecular, would be the key to overcoming endogenous resistance potential.

The antibacterial R&D business is in the midst of an unglamorous comeback born from medical necessity as well as from the steady rise of antibiotic-resistant pathogens that dictates a need for new products. New versions of familiar antibacterials are expected to keep the worst bacterial pathogens at bay until something truly amazing comes along to deliver a more substantial setback to the pathogens.
