Initial and steady-state pharmacokinetics of a vaginally administered formulation of progesterone
Article Abstract:
Vaginal suppositories may be a convenient way of administering progesterone. Injection is painful and must be done daily. Oral administration requires large doses two or more times per day. Ten healthy postmenopausal women inserted a vaginal suppository once daily for 14 days. Menopausal women were used so that internal production of progesterone would not confuse the results. Two different doses were used. Blood samples were drawn periodically and analyzed for progesterone. Progesterone reached its maximum concentration within an average of three hours of insertion. Lower doses were required to reach therapeutic levels than with oral progesterone. The larger dose did not result in higher blood levels, suggesting a maximal absorption rate by vaginal tissues. Progesterone levels varied markedly within individual women from day to day.

Misoprostol versus methylergometrine: Pharmacokinetics in human milk
Article Abstract:
A study is conducted to compare breast milk pharmacokinetics between misoprostol 200 mug and methylergometrine 250 mug after single oral dosing in women who require postpartum uterotonic therapy. The results show that misoprostol enters and leaves breast milk at twice the rate of methylergometrine and has one-third the milk/plasma ratio, thus substantially lowering infant exposure to the drug.
