Abnormalities in the regulation of variable region genes that encode for antibodies to DNA may be a central factor in the pathogenesis of systemic lupus erythematosus
Article Abstract:
Patients with systemic lupus erythematosus (SLE) may inherit certain types of genes that cause increased production of antibodies against their own DNA. SLE is a chronic inflammatory disorder that affects the skin, joints, kidneys and connective tissues. The role of DNA antibodies in the development of SLE has been observed both in animals and humans. Variable region genes (V genes) contain the genetic code from which DNA antibodies are produced. Some individuals may inherit individual autoreactive germline V genes that increase the risk of developing SLE. These genes may be responsible for the increased production of DNA antibodies in SLE patients. Some individuals who inherit autoreactive V genes may not develop SLE. Certain regulatory factors may prevent increased production of DNA antibodies from autoreactive V genes in these individuals.

Is immunogenetic susceptibility to neuropsychiatric systemic lupus erythematosus (SLE) different from non-neuropsychiatric SLE?
Article Abstract:
Specific HLA subtypes appear to be frequently associated with the psychiatric features of systemic lupus erythematosus (SLE). The HLA system is part of the immune system that codes for tissue compatibility. HLA subtypes were determined for 93 patients with SLE including 42 with neuropsychiatric lupus (NPSLE) and 191 healthy blood donors (control group). Patients with NPSLE carried HLA-DQ2 and HLA-DR9 subtypes more frequently and HLA-DR4 less frequently than those with SLE. Patients with SLE carried the HLA-DR3 subtype more frequently than the control group.
