The effect of recombinant erythropoietin on intracellular free calcium in erythropoietin-responsive cells
Article Abstract:
Erythropoietin is a hormone which works directly on the marrow to increase the rate of red blood cell production. The role of intracellular free calcium in this process is investigated in this experiment. The experiment used cells responsive to erythropoietin that were collected from the spleens of mice which had been previously treated with phenylhydrazine. The level of calcium in the cells was measured and the cell suspensions were treated with a calcium ionophore, which allows calcium to pass more easily through cell membranes. This raised the level of intracellular calcium by 90 percent and it was found that the increased calcium content had no effect on the number and association of the cells. When bone marrow cell colonies were treated with a chemical which removes calcium, growth was inhibited. This effect may have resulted from toxic effects of the chemical treatment, because replacement of calcium ions failed to restore colony proliferation. The role of erythropoietin in the development of red blood cells does not appear to be affected by changes in calcium ion level.

Expression of protooncogene c-myb in normal human hematopoietic cells
Article Abstract:
Many human cells contain segments of genetic information that are related to genes which become active in the production of neoplasms (cancers). These segments, called protooncogenes, are probably involved in the mechanism of cell development and differentiation (the process which converts a generalized cell into a muscle cell or a cell of the skin, etc.). This study examines the expression of the specific oncogene, c-myb, by studying the surface proteins (antigens) that mark the different types of blood cells. Certain lines of marrow cells, particularly the CO34+ lymphocyte, showed a high level of c-myb expression. Nearly all bone marrow cells were found to contain at least some level of c-myb, and this may relate to the process of proliferation and differentiation.
