Modulation of platelet aggregation by an L-arginine-nitric oxide pathway
Article Abstract:
L-arginine and nitric oxide are found to inhibit collagen-mediated blood platelet aggregation resulting in an increase in intraplatelet cGMP. This reaction is stimulated by a sudden rise in intracellular Ca2+ which leads to aggregation. The Ca2+-initiated activity generates nitric oxide from L-arginine via nitric oxide synthase reaction. The formation of nitric oxide regulates soluble guanylyl cyclase which eventually reduces the aggregation process. Moreover, agents that interfere with nitric oxide formation or limit its effects enhance the occurrence of platelet aggregation.

Nitric oxide has indisputably been demonstrated to be an important biological entity. Among its other functions, nitric oxide has clearly been shown to act as a significant regulator of general cellular processes such as gene expression and mitochondrial function. The NOS structure continues to conceal several surprising discoveries. For example, X-ray diffraction analysis at 0.19 nm resolution has shown that eNOS is a metalloenzyme able to bind ZN with high affinity.

Nitric oxide takes center-stage with newly defined roles
Article Abstract:
Studies show that nitric oxide may have potential applications in clinical medicine. Nitric oxide, which is synthesized from L-arginine by nitric oxide synthase, binds with guanylyl cyclase to activate cardiovascular and neural actions. It can also be added to intact cells to cause nitrosylation of nucleic acids and break DNA strands. Other studies also provide evidences supporting the possible role of nitric oxide in the immune system.
