A whole genome approach to in vivo DNA-protein interactions in E. coli
Article Abstract:
An enzymatic technique for efficiently determining which parts of DNA sequences are bound by proteins in vivo uses a whole-genome approach. Those genomic targets not protected in vivo by protein factors which interfere with methylase action are methylated by artificially introduced methylases. Those which are protected in vivo remain unmethylated, and are then detected using restriction endonucleases which are sensitive to methylation. This technique is less damaging to the cell than other in vivo methods and makes fewer assumptions about protein-binding sites.

A full understanding of the large or rapid climatic changes associated with ice ages over the past 2.5 million years would help to assess future climatic changes. Rohling and colleagues have estimated sea-level lows of the past 500,00 years and have calculated that sea level was around 140 m during an ice age known as MIS12, that peaked around 450,000 years ago, 140 m lower than today.
