Bacterial DNA causes septic shock
Article Abstract:
Bacterial DNA may play a role in triggering septic shock during Gram-positive or Gram-negative bacterial infections. This potential role was investigated by examining whether DNA prepared from Gram-positive or Gram-negative bacteria or active synthetic oligonucleotides prompt rapid tumor-necrosis-factor-alpha (TNF-alpha) production in macrophages to cause toxic shock in D-galactosamine-sensitized mice. Results showed that mice injected with Gram-positive DNA, Gram-negative DNA and active synthetic oligonucleotides exhibited high but transient serum concentrations of TNF-alpha, which triggered apoptotic cell death and led to toxic shock due to acute liver failure.

Critical role of TRAF3 in the toll-like receptor-dependent and -independent antiviral response
Article Abstract:
TRAF3 associates with the TLR adaptors TRIF and IRAKI, as well as downstream IRF3/7 kinases TBK1 and IKK-epsiv suggests that TRAF3 serves as a critical link between TLR adaptors and downstream regulatory kinases important for IRF activation. TTRAF3 is an important component of TLR-independent viral recognition pathways, which demonstrates that TRAF3 is a major regulator of type I IFN production and the innate antiviral response.

Specificity in toll-like receptor signaling through distinct effector functions of TRAF3 and TRAF6
Article Abstract:
TRAF3 deficient cells overproduce pro-inflammatory cytokines owing to defective IL-10 production and despite their structural similarity, the functions of TRAF3 and TRAF6 are largely distinct. TRAF3 is also recruited to the adaptor TRIF and is required for marshalling the protein kinase TBK1 into TIR signaling complexes, thereby explaining its unique role in activation of the IFN response.
