Article Abstract:
The progress of research on endothelins is discussed. Endothelins are highly active peptides which cause elevation of blood pressure via vasoconstriction. They also influence other physiological processes such as electrolyte balance, cell proliferation and neurotransmitter release. Progress has been made in identifying and characterizing endothelin receptors and in the development of potent and orally active receptor antagonists. However, the endothelin-converting enzyme has yet to be identified. The implications of endothelin research for the treatment of high blood pressure, congestive heart failure and hypotension are discussed.

Endothelin versatility
Article Abstract:
The naturally occurring endothelins, ET-1, ET-2 and ET-3, are important in conducting pathophysiological activities, constriction of blood vessels, expressing in non-vascular tissues and rise in blood pressure in mammals. In rats, endothelin is responsible for monocrotaline-induced pulmonary hypertension. The growth of neural-crest-derived tissues is due to the action of ET-1 on endothelin A receptors. For the development of neural-crest-derived melanocytes and enteric neurons combined action of ET-3 and endothelin B receptors seems essential.

Inhibition of myocardial endothelin pathway improves long-term survival in heart failure
Article Abstract:
Long-term treatment with an endothelin-receptor antagonist leads to inhibition of the myocardial endothelin pathway. This improves the chances of long-term survival in cases of heart failure. A significant improvement in the left ventricular dysfunction, and inhibition of ventricular remodelling follows the beneficial effect of the myocardial endothelin pathway inhibition. The levels of endothelin-1 mRNA are high in the cardiac myocytes than in the coronary endothelium of rats surviving heart failure.
