Microglial activation by Alzheimer amyloid precursor protein and modulation by apolipoprotein E
Article Abstract:
Researchers have investigated whether a secreted derivative of beta-amyloid precursor protein (beta-APP) (sAPP-alpha) can bring about inflammatory reaction in microglia, brain cells of monocytic lineage. This involved treating the N9 microglial cell line with sAPP and measuring the activity of the transcription factor NF-kappa-B by electrophoretic mobility-shift assay (EMSA). It was established that sAPP can activate microglia, boosting their neurotoxicity.

Alzheimer amyloid protein precursor complexes with brain GTP-binding protein G-O
Article Abstract:
Amyloid precursor protein (APP), which gives rise to the amyloid beta/A4 protein known to form tissue deposits in the brains of patients with Alzheimer's disease, builds a complex by acting as a receptor to G-O, a GTP-binding protein in the brain. GTP-gamma-S caused APP to fail to react to G-O. It is therefore feasible that Alzheimer's disease results from the malfunctioning of the APP-G-O system produced by structural changes in APP.
