The TAK1-NLK-MAPK-related pathway antagonizes signalling between beta-catenin and transcription factor TCF
Article Abstract:
An analysis of the impact of TAK1 on the transcriptional activities of a beta-catenin-T-cell factor (TCF) complex using two luciferase reporter constructs has given an insight into whether TAK1 can affect Wnt signal transduction. The reporter constructs contained either multimeric TCF-binding sites or mutated TCF-binding sites. It was established that regulation of TCF as both an activate and a repressor could reflect a balance between TCF with and without beta-catenin. There is conservation between mammals and C. elegans in the downregulation of HMG-domain proteins through MAPK-like pathways.

The kinase TAK1 can activate the NIK-I-kappa-B as well as the MAP kinase cascade in the IL-1 signalling pathway
Article Abstract:
The MAPKK kinase TAK1 has been found to act upstream of NF-kappa-B-inducing kinase (NIK) in the interleukin-1-activated signalling pathway. Immunoblot analysis of NIK from cells overexpressing wild type NIK indicated that NIK migrated as a doublet, possibly because of autophosphorylation and/or phosphorylation by activated I-kappa-B kinase-alpha. TAK1 has been identified as the kinase that connects the protein TRAF6 with the NIK-IKK cascade in the Il-1 signalling pathway.

MAP kinase and Wnt pathways converge to downregulate an HMG-domain repressor in Caenorhabditis elegans
Article Abstract:
The apparently different outputs of Wnt signalling can be explained by the fact that Wnt signalling stabilizes beta-catenin in all cases. It has been established that a mitogen-activated protein kinase-related pathway cooperates with Wnt signal transduction to downregulate POP-1 activity. The genes mom-4 and lit-1 are needed to downregulate POP-1, both in the posterior daughter cell E and in other posterior daughter cells.
