Issues are presented concerning the functioning of synaptic terminals which prime vesicles at release sites for exocytosis and then re-supply vesicles to the release sites.

Transport, docking and exocytosis of single secretory granules in live chromaffin cells
Article Abstract:
It is possible to use evanescent-wave fluorescence microscopy to observe single secretory granules undergoing exocytosis. These granules leave behind patches of bare plasmalemma. It seems that granules reach the plasmalemma in a directed way. Once they have arrived, they lose lateral motility, and it appears that they bind to the plasmalemma. Future research must look at the motors which carry granules to their docking sites.

The exocytotic event in chromaffin cells revealed by patch amperometry
Article Abstract:
It is possible to use cell-attached capacitance measurements linked with electrochemical detection of catecholamines to analyze exocytosis of individual chromaffin granules. It was established that the fusion-pore diameter remains at <3 nm for a period of variable length, sometimes up to several seconds, before expanding. In single chromaffin cells all granules contain around the same concentration of catecholamines, irrespective of size. The amperometric foot reflects release of around 2.5 x 10(super 7) molecules per second through fusion pores with conductances of around 500 pS.
