A new sensitive, whole-cell hybridization technique for detection of bacteria involving a biotinylated oligonucleotide probe targeting rRNA and tyramide signal amplification
Article Abstract:
The sensitivity of fluorescent in situ hybridization can be improved with a new tyramide signal amplification (TSA) method using biotinylated oligonucleotide probes and streptavidin-horseradish peroxidase. There was a 7- to 12-fold amplification observed when the method was used in studying the starved cells of Salmonella typhimurium and Deleya aquamarina. The method has the highest sensitivity among signal amplification systems using single rRNA sequences.

Comparison of blue nucleic acid dyes for flow cytometric enumeration of bacteria in aquatic systems
Article Abstract:
Eight blue nucleic acid dyes were compared on their utility in staining living and dead bacteria from freshwater and seawater samples undergoing flow cytometric analysis for bacterial load. Results reveal that the SYBR-II and SYTO-9 are efficient in staining living and dead microorganisms from freshwater samples. Bacterial enumeration from fixed and unfixed seawater samples are better carried out with the SYBR-II dye.

Diversity of Salmonella strains isolated from the aquatic environment as determined by serotyping and amplification of the ribosomal DNA spacer regions
Article Abstract:
Researchers used conventional serotyping and the ribosomal spacer-heteroduplex polymorphism method to identify more than 40 different Salmonella serotypes in the Tech River watershed on the south coast of France. Some serotypes were only found after storms and could represent contamination from farms.
