A simple method for quantification of uncultured microorganisms in the environment based on in vitro transcription of 16S rRNA
Article Abstract:
A novel method for quantitative hybridization was developed for the quantification of uncultured microorganisms based on the generation of nucleic acid standards. The method involves the quantitative estimate of a species added at different concentrations to an artificially assembled community based on ribosomal ribonucleic acids (rRNA) transcribed from cloned templates. Furthermore, the in vitro-transcribed rRNA can be utilized as a standard in quantitative probing of a microbial community's nucleic acids.

Bias in template-to-product ratios in multitemplate PCR
Article Abstract:
The potential causes and the extent of bias in polymerase chain reaction amplification of 16S ribosomal DNAs were investigated. Quantification and comparison of template and product ratios showed that there was considerable and reproducible overamplification of specific templates. Results indicated that bias may be reduced considerably by using high template concentrations, by performing fewer cycles and by mixing replicate reaction preparations.

Two new Mycobacterium strains and their role in toluene degradation in a contaminated stream
Article Abstract:
Two toluene-degrading Mycobacteria strains are isolated from a toluene-contaminated freshwater stream. The two strains have different rates of toluene degradation. Morphological, biochemical and genetic analysis of the isolated strains confirmed their relationship to the genus Mycobacteria. These fast-growing strains are closely related to Mycobacterium aurum and M. komossense based on the analysis of their 16S ribosomal RNA sequences.
