Autocrine growth and anchorage independence: two complementing Jun-controlled genetic programs of cellular transformation
Article Abstract:
The viral form of the AP-1 subunit Jun sets up one aspect of transformation in chick embryo fibroblasts (CEFs). The complexity of AP-1 factors has been cut down by constructing Jun bZip mutants that can dimerize and transactivate efficiently with no more than a restricted set of partner subunits. The object was to identify programs that relate to cell transformation. A Jun mutant selective for the Fos family induced anchorage-independent growth but no growth-factor independence when introduced into CEFs. A c-Jun mutant with preference for ATF2-like proteins caused growth-factor independence but no growth in agar. Coexpressed, the mutants reestablished the combined transformation program.

A mutation is introduced into the mouse thyroid hormone receptor alpha1 gene that lowers affinity to thyroid hormone 10-fold to determine the role of unliganded thyroid hormone receptor alpha1 in neuronal tissues. The resulting heterozygous mice exhibit several distinct neurological abnormalities, extreme anxiety, reduced recognition memory, and locomotor dysfunction.

Article Abstract:
Glucocorticoid receptor (GR) -mediated transrepression factors AP-1 and NF-(kappa)B, responsible for most of the anti-inflammatory effects of glucocorticoid, are initiated by the tethering of GR to the promoters of target genes. The report that this tethering is mediated by a nuclear isoform of the focal adhesion LIM domain protein Trip6 is presented.
