Characterization of an insertion sequence, IS12528, from Gluconobacter suboxydans
Article Abstract:
Mutants with reduced alcohol dehydrogenase (ADH) activity were analyzed to determine the role of spontaneous mutations in promoting Gluconobacter spp. ethanol oxidation deficiency. Analysis of the IFO12528 genes which encodes the membrane-bound ADH in Gluconobacter spp. indicated the presence of a new insertion sequence named IS12528. The novel insertional sequence promotes the inactivation of the g3-adhA-g3-adhB gene cluster. Furthermore, the transposable element is responsible for ethanol oxidation deficiency in Gluconobacter spp.

A new insertion sequence IS1452 from Acetobacter pasteurianus
Article Abstract:
A new insertion sequence IS1452 can cause spontaneous mutations at high frequencies in strains of Acetobacter affecting acetic acid resistance, ethanol oxidation, and cellulose production. This element inserts in and disrupts the alcohol dehydrogenase S gene. Replicative transposition was observed for IS1452 since it is accompanied by an increase in the copy number of the transposon. The frequent transposition of these transposons suggests that these indigenous IS elements are responsible for genetic instability in Acetobacter.

Characterization of the genes encoding the three-component membrane-bound alcohol dehydrogenase from Gluconobacter suboxydans and their expression in Acetobacter pasteurianus
Article Abstract:
The primary structures of the three subunits of Gluconobacter suboxydans IFO12528 alcohol dehydrogenase (g3-ADH) were analyzed based on the nucleotide sequences of the cloned genes. Analysis of the primary translation products deduced from the nucleotide sequences and NH2-terminal amino acid sequences of purified g3-ADH indicated the presence of a typical signal sequence in their NH2-termini. Furthermore, the AdhS subunit is required for ADH activity in Acetobacter pasteurianus.
