Redox chemistry in laccase-catalyzed oxidation of N-hydroxy compounds
Article Abstract:
Redox chemistry is discussed relative to laccase-catalyzed oxidation of N-hydroxy compounds. Three N-OH compounds that can mediate a range of laccase-catalyzed biotransformations have been studied. The mechanisms of the enzymatic oxidations were studied with seven fungal laccases. It is probable that an outer-sphere sort of single-electron transfer from the substrate to laccase and proton release is involved in the rate-limiting step of N-OH oxidation.

Purification, characterization, molecular cloning, and expression of two laccase genes from the white rot basidiomycete Trametes villosa
Article Abstract:
Two laccase genes from the white rot basidiomycete Trametes villosa have been purified, characterized, molecularly cloned and heterologously expressed. The purified enzymes were found to share several properties with laccases found in other Trametes species. Data from the Southern blot probed with the LCC cDNA insert indicated the presence of more than two laccase genes in Trametes villosa.

Characterization of the gene encoding an extracellular laccase of Myceliophthora thermophila and analysis of the recombinant enzyme expressed in Aspergillus oryzae
Article Abstract:
A DNA segment containing an extracellular laccase (MtL) from the fungus Myceliophthora thermophila was cloned and expressed in Aspergillus oryzae. Its amino acid sequence has been found to be similar to that of other fungus species especially the Sphaeriales. The recombinant MtL was synthesized as a preproenzyme. It has a molecular mass of approximately 100 to 140 kDA and contains 40% to 60% glycosylation.
