The zinc ribbon domains of the general transcription factors TFIIB and Brf: conserved functional surfaces but different roles in transcription initiation
Article Abstract:
The zinc ribbon domains of the general transcription factors Brf and TFIIB are discussed relative to conserved functional surfaces and different roles in transcription initiation. The function of the conserved zinc-binding domains in the related Po lI- and Pol III-specific factors, TFIIB and Brf, was studied using 3-D structure modeling and mutagenesis studies. One conserved surface of the zinc ribbon, mostly in the second and third antiparallel beta strands, has two functions.

Recruitment of the transcriptional machinery through GAL11P: structure and interactions of the GAL4 dimerization domain
Article Abstract:
The binding of GAL4 to GAL11P in Saccharomyces cerevisiae is a unique structural motif for an activating region able to bind to one target to cause gene expression, although the binding is an artificial interaction. GAL4-dd is made up of three helical segments connected by flexible loops. Dimerization is via coiled-coil interactions of the three helices. Nuclear magnetic resonance titration was used to study interaction of the GAL4-dd with GAL11P(sub.261-352). GAL11P is a part of the RNA-polymerase II holoenzyme.

Recruitment of the SWI/SNF chromatin remodeling complex by transcriptional activators
Article Abstract:
The SWI/SNF chromatin remodeling complex and its recruitment by transcriptional activators are discussed. An activation domain is required. REcruitment is not dependent on promoter sequences, TBP, or RNA polymerase II holoenzyme. Acidic activation domains can target SWI/SNF remodeling activity and SWI/SNF activity can be targeted by gene-specific activators. The recruitment can be independent of Pol II holoenzyme.
