Defective DNA-dependent protein kinase activity is linked to V(D)J recombination and DNA repair defects associated with the murine scid mutation
Article Abstract:
A study of the defective protein kinase activity that depends on DNA reveals that the activity is absent in murine cells that are homozygous for the severe combined immune deficiency mutation (scid) and V3 mutant hamster cells, due to defects in DNA-PK(cs). DNA repair and recombination defects of V3 cells are compensated by yeast artificial chromosome with the DNA-PK(cs). The XRCC7 gene codes for DNA-PK(cs) that is stimulated by its binding to DNA ends.

DNA-dependent protein kinase catalytic subunit: a relative of phosphatidylinositol 3-kinase and the ataxia telangiectasia gene product
Article Abstract:
The cloning of a series of DNA-dependent protein kinases (DNA-PKcs) reveals that they belong to the phosphatidylinositol (PI) 3-kinase family. DNA-PK is capable of phosphorylating proteins but has no action over lipids. The similarity to PI kinases is visible in the cell cycle control, DNA repair and DNA damage activities. DNA-PK is a serine/threonine protein kinase that occurs in a range of eukaryotes and takes part in DNA-double stranded break repair.

The DNA-dependent protein kinase: requirement for DNA ends and association with Ku antigen
Article Abstract:
The Ku antigen is a heterodimeric protein complex present in various mammalian systems whose biological function remains unclear. Experiments on the DNA binding specificity of DNA-dependent protein kinase (DNA-PK) showed that the enzyme binds to DNA ends and requires them for activity. Biochemical and immunological experiments showed that the DNA-binding component of DNA-PK is the Ku antigen.
