Deletion of the H19 differentially methylated domain results in loss of imprinted expression of H19 and Igf2
Article Abstract:
H19 and Igf2 imprinting require differentially methylated sequences of H19. Mice with a 1.6-kb targeted deletion of the differentially methylated domain (DMD) have been studied for allelic expression of H19 and Igf2, a linked but oppositely imprinted gene. When the DMD deletion is inherited paternally, H19 is activated and expression of Igf2 is diminished. Maternal transmission of the mutation is associated with reduced expression of H19 and activation of Igf2.
