Detection and enumeration of a tagged Pseudomonas fluorescens strain by using soil with markers associated with an engineered catabolic pathway
Article Abstract:
Mannopine (MOP) cyclase, that catalyzes the catabolism of MOP, can be used as a marker for the detection and direct selection of genetically engineered Pseudomonas fluorescens present in soil samples. MOP cyclase allows the use of a nonselective medium to retrieve culturable microbes, and identify and enumerate them using colony immunoblot assays. MOP cyclase is useful as a marker even for microorganisms that are incapable of utilizing agropine as a sole carbon source.

Conjugal transfer but not quorum-dependent tra gene induction of pTiC58 requires a solid surface
Article Abstract:
The tra genes of both the nopaline-type Ti plasmid pTiC58 and wild-type Ti plasmids of Agrobacterium tumefaciens can be induced and the conjugal transfer mechanism can be expressed in liquid environments. However, a solid stratum is necessary for the formation of stable mating pairs and subsequent transfer of the Ti plasmid. Nonetheless, the regulatory system can stimulate tra gene expression to equivalent levels not only on solid surfaces but also in liquid.

Involvement of nitrate reductase and pyoverdine in competitiveness of Pseudomonas fluorescens strain C7R12 in soil
Article Abstract:
Both nitrate reductase and pyoverdine contribute to the competitiveness of Pseudomonas fluorescens strain C7R12, although they contribute more to saprophytic ability than rhizosphere competence. The advantage conferred by nitrate reductase is greatest at low oxygen levels.
