Development and application of 16S rRNA-targeted probes for detection of iron- and manganese-oxidizing sheathed bacteria in environmental samples
Article Abstract:
Sequencing of 16S rRNA genes of Leptothrix and Sphaerotilus strains leads to an oligonucleotide probe that helps in the detection of iron and manganese oxidizing sheathed filamentous bacteria in a wetland habitat. Positive signals to the probes are obtained from the sheathed filamentous bacteria, some of which are morphologically similar to Leptothrix discophora. Unknown filamentous sheathed bacteria are also identified, which are morphologically similar to Sphaerotilus natans.

Development of a robust flow cytometric assay for determining numbers of viable bacteria
Article Abstract:
The fluorescent anionic membrane potential probe bis-(1,3-dibutylbarbituric acid)trimethine oxonol is used as a robust indicator of bacterial cell viability, by flow cytometry. The live and dead cells in a population are detected and counted 10 minutes after sampling. The flow cytometry technique is indispensable to identify physiological changes of each microorganism because of external perturbations.

Physical characterization and quantification of bacteria by sedimentation field-flow fractionation
Article Abstract:
A new technique of field-flow fractionation enumerates bacteria and determines their density, size and mass by using cultures of common bacterial species as models. The new method estimates bacterial biomass directly and has advantages over other methods for calculating biomass. The sdFFF method should make it possible to determine biomass accurately at a large number of stations.
