Development and evaluation of functional gene arrays for detection of selected genes in the environment
Article Abstract:
Glass-based microarray hybridization can be used to determine microbial gene composition and diversity, according to researchers who use it to study genes involved in nitrogen cycling from pure cultures and cultures isolated from marine sediments. However, the technique is less useful for measuring the abundance of specific genes in environmental samples.

Evaluation of PCR-generated chimeras, mutations, and heteroduplexes with 16S rRNA gene-based cloning
Article Abstract:
PCR-generated artifacts can be reduced during 16S rRNA gene-based analysis of microbial communities by using AmpliTaq and fewer than 20 PCR cycles. PCR-generated heteroduplexes can be removed by polyacrylamide gel purification or T7 endonuclease I digestion.

Pseudomonas stutzeri nitrite reductase gene abundance in environmental samples measured by real-time PCR
Article Abstract:
Real-time PCR analysis of the denitrifying nitrite reductase gene nirS shows that Pseudomonas stutzeri is not the dominant denitrifier in marine sediments. However, the organism is common in lake sediment and groundwater.
