Differentiation of Vibrio alginolyticus strains isolated from Sardinian waters by ribotyping and a new rapid PCR fingerprinting method
Article Abstract:
A proposed polymerase chain reaction fingerprinting technique for the differentiation of 30 Vibrio alginolyticus strains isolated in Sardinian waters has been found to possess superior discriminative power. Conserved and specific internal sequences belonging to the IS256 family were amplified and analyzed by a computer program to establish genetic relationships, after which results were compared with patterns obtained by ribotyping with HindIII, KpnI, and XbaI restriction enzymes. The novel method was found to be able to differentiate the strains analyzed into 12 different patterns, while ribotyping with XbaI, which generated the most number of patterns, yielded only seven different profiles.

Identification and quantification of toxic chemicals by use of Escherichia coli carrying lux genes fused to stress promoters
Article Abstract:
The luxCDABE bioluminescence genes of the Vibrio fischeri lux system were employed for the qualitative and quantitative determination of toxic chemicals depending on their capability to activate certain Escherichia coli promoters. The biological method may be applied for prescreening toxic agents in environmental samples and downstream procedures, and prescreening of biological and fermentation processes for sudden changes in chemical composition.

Effects of temperature and salinity on Vibrio vulnificus population dynamics as assessed by quantitative PCR
Article Abstract:
The population dynamics of Vibrio vulnificus and its relationship to the physicochemical condition of Barnegat Bay using a most-probable-number (MPN)-PCR approach for cell enumeration is presented. The relationship between salinity and V. vulnificus abundance is dependent on salinity regime or not and the temperature-mediated population decline is due to entry of the population into the viable but nonculturable (VBNC) state are explained.
