Targeting of Ikaros to pericentromeric heterochromatin by direct DNA binding
Article Abstract:
Targeting of Ikaros, a sequence-specific DNA-binding protein essential for lymphocyte development, to pericentromeric heterochromatin is discussed. It is achieved by direct DNA binding. Results may be consistent with presence of Ikaros binding sites within centromeric DNA repeats.

Research findings indicate that Ikaros, a regulator of lymphopoiesis, has a direct role in heritable gene inactivation. Mutations that selectively disrupt Ikaros binding to an integrated gene-encoding terminal transferase (TdT) promoter stopped down-regulation on differentiation.

Helios, a T cell-restricted Ikaros family member that quantitatively associates with Ikaros at centromeric heterochromatin
Article Abstract:
Helios, the p70 gene, is a T cell-restricted Ikaros family member. It associates with Ikaros isoforms without discrimination when overexpressed and associates quantitatively with Ikaros at centromeric heterochromatin. Ikaros is in most hematopoietic cells. Only a fraction of the Ikaros, likely that associated with Helios, localizes centromerically in T cells. It seems Helios is a limiting regulatory subunit for Ikaros within centromeric heterochromatin. Immunoaffinity chromatography is useful in identifying Ikaros partners.
