Evaluation of bottlenecks in the late stages of protein secretion in Bacillus subtilis
Article Abstract:
A study revealed that only five potential bottlenecks in the secretion pathway of Bacillus subtilis have been documented. It is believed that heterologous proteins may form insoluble aggregates within the cytoplasm due to the limited activity of microbial chaperones. Since secretion bottlenecks relating to the secreted protein have been poorly identified, the bottlenecks of four different heterologous reporter proteins from eubacteria and eukaryotes, and other substances with similar gene expressions and secretion signals were compared.

Use of alkaline phosphatase as a reporter polypeptide to study the role of the subtilin leader segment and the SpaT transporter in the posttranslational modifications and secretion of subtilin in Bacillus subtilis 168
Article Abstract:
Alkaline phosphatase was employed as a reporter polypeptide for the examinination of the SpaT transporter and the subtilin leader segment in Bacillus subtilis 168 posttranslational modifications and subtilin secretion. The subtilin leader segment's proteolytic cleavage was disrupted by a large in-frame deletion from the SpaT central region. Enzymatic acticity and amino acid composition analysis also reveal that the subtilin leader segment's secreted AP did not undergo posttranslational modification.

Direct selection of cloned DNA in Bacillus subtilis based on sucrose-induced lethality
Article Abstract:
Sucrose-induced lethality due to the inactivation of sacB gene by the insertion of heterologous DNA helps to select for cloned DNA in Bacillus subtilis. The W29 mutation inhibits growth of B. subtilis in a sucrose containing medium. The inactivation of fructose polymerase encoded by sacB reveals that levan production is essential for the sucrose-induced lethality. The mutation has no effect on B. subtilis in the absence of sucrose.
