GcvA binding site 1 in the gcvTHP promoter of Escherichia coli is required for Gcva-mediated repression but not for Gcva-mediated activation

Research demonstrates that the operon gcvTHP responsible for encoding glycine cleavage enzyme system is negatively regulated by site 1 binding site of gcvTHP control region comprising the base region from 34 to 69 as shown by base substitution and mutational studies.

GcvR interacts with GcvA to inhibit activation of the Escherichia coli glycine cleavage operon
Article Abstract:
Results show that in Escherichia coli GcvA gene regulates the glycine cleavage enzyme system operon gcvTHP positively and negatively in combination with GcvR. Data indicate that repression of gcvTHP operon expression by direct interaction between GcvR and GcvA in vivo is mediated by the C-teminal half of GcvA.

The Escherichia coli glycine transport system and its role in the regulation of the glycine cleavage enzyme system
Article Abstract:
A serine biosynthesis (serA)- and glycine transport-disrupted Escherichia coli K12 mutant has a glycine cleavage negative (GCV-) phenotype with lower levels of glycine uptake. Exogenous glycine level increase induces similar enhancements in the induction and uptake in both the mutant and wild-type strains. gcvT:lacZ fusion activation is similar in the lycA and wild type strains that are grown in a tripeptide or threonine medium with glycine. The introduction of a gcv mutation into the cycA strain enhances activation of gcvT::lacZ fusion, indicating that cyc regulates gcv through the transportation of glycine into the cell, activating gcv expression.
