High-throughput screen for poly-3-hydroxybutyrate in Escherichia coli and Synechocystis sp. strain PCC6803
Article Abstract:
The development of two novel methods for detecting poly-3-hydroxybutyrate (PHB) based on Nile red fluorescence that are both quantitative and maintain cell viability is described. The straining protocols are novel techniques that are useful in the high-throughput evaluation of combinatorial libraries of Synechocystis and Escherichia coli by using fluorescence-activated cell sorting to identify mutants with increased PHB-accumulating properties.

A transposition-induced mutant of Nostoc ellipsosporum implicates an arginine-biosynthetic gene in the formation of cyanophycin granules and of functional heterocysts and akinetes
Article Abstract:
An experiment was conducted to analyze a transposon mutant NE1 of Tn5-1058-mutagenized Nostoc ellipsosporum. The N. ellipsosporum was characterized by abnormal akinetes and heterocysts as well as inadequate cyanophycin granules. Test results revealed the formation of cyanophycin granules due to the combination of akinetes and nitrate. In addition, the mutation of a newly identified gene was observed.
