How do highly branched (colonial) mutants of Fusarium graminearum A3/5 arise during Quorn myco-protein fermentations?
Article Abstract:
The nuclear distribution of heterokaryons in Fusarium graminearum is studied. The possibility of mycelia with colonial mutant phenotypes arising in the process of fermenting Quorn microprotein for human consumption following fragmentation of heterokaryotic mycelia is determined. Results indicate three factors behind the appearance of highly branched colonial mutants in cultures of F. graminearum. Moreover, a strategy that uses a heterokaryon formed between auxotrophic mutants to prevent the appearance of colonial mutants in Quorn mycoprotein fermentations is not feasible.

Study of the glucoamylase promoter in Aspergillus niger using green fluorescent protein
Article Abstract:
An Aspergillus niger strain expressing a red-shifted green fluorescent protein (GFP) in the cytoplasm regulated by the glucoamylase promoter (PglaA) was characterized with regards to its physiology and morphology. Red-shifted GFP was first produced after five hrs induction. The strain was characterized in glucose-restricted continuous cultures. The area of the mycelium stained with cytoplasmic GFP was found to increase with increasing specific growth rate, showing that GFP can be employed as a marker of cellular activity in this type of cultivation.
