Identification and characterization of an IkB kinase
Article Abstract:
The NF-kB-inducing kinase (NIK) is required in the activation of the transcription factor NF-kB by tumor necrosis factor and interleukin-1. A protein kinase previously known as CHUK was identified in a yeast two-hybrid screen for NIK-interacting proteins. A NF-kB-dependent reporter gene is activated by overexpression of CHUK. Moreover, CHUK phosphorylates IkB-alpha on both serine 32 and 36, which is enhanced by NIK costimulation. The findings indicate that CHUK is an NIK-activated IkB-alpha kinase.

Tumor necrosis factor's cytotoxic activity is signaled by the p55 TNF receptor
Article Abstract:
Tumor necrosis factor (TNF) is a potent cytokine produced by macrophages. It causes hemorrhagic necrosis of transplanted tumors, cytotoxicity, plays a role in endotoxic shock and other biological processes. Two TNF receptors, a 55 kd receptor and a 75 kd receptor, have been characterized in both human and mouse cDNAs. The cytotoxic effects of agonist anti-p55 and anti-p70 antibodies were assayed and the p55 recptor was found to signal TNF cytotoxicity in contrast to p70.

The TNFR2-TRAF signaling complex contains two novel proteins related to baculoviral inhibitor of apoptosis proteins
Article Abstract:
The c-inhibitor of apoptosis (IAP) 1 and c-IAP2 proteins are intracellular signal transducers involved in signal transduction by the tumor necrosis factor receptor 2 (TNFR2). The proteins are members of the baculovirus IAP family. They bind to the TNFR-associated factor 1 (TRAF1) and TRAF2 by the N-terminal of the baculovirus IAP repeats. The involvement of c-IAP1 and c-IAP2 in the signaling pathway requires the presence of a TRAF2-TRAF1 heterocomplex.
